Advanced ingredients, delivery and quality standards
Research date: 2026-11-05. Doses are for an 18 kg (40 lb) dog. Grades: A randomized placebo-controlled canine trial on the target outcome; B controlled canine trial but small, sponsor-run, biomarker-only or a combination product; C open-label or pilot in dogs; none no canine data. Bulk prices are marketplace asking prices, not quotes.
What moves the formula
| Add | Line | Grade | Dose (40 lb) | Why it earns a place |
|---|---|---|---|---|
| Eggshell membrane (NEM) | Mobility | B | 13.5 mg/kg = about 245 mg | Randomized placebo-controlled canine trial (Ruff 2016, n=51): pain and function improved at week 1, cartilage-breakdown marker CTX-II down 48%. A 2025 Virbac trial (n=46) confirmed pain-interference benefit. Nobody in the CBD category uses it. Egg allergen. |
| KSM-66 ashwagandha | Chill | A (single, sponsor-linked) | 15 mg/kg = about 270 mg | Kaur 2022 (n=24, 4 weeks): urine cortisol down, fear and anxiety scores down, pain scores down, no adverse events. Two later geriatric-beagle studies. Root extract, 5% withanolides. Mild sedation may add to CBD. |
| Alpha-casozepine (Lactium / Zylkene) | Chill | A/B | 15 mg/kg = about 270 mg | Non-inferior to selegiline (Beata 2007), a 2026 placebo crossover, and the only botanical in a canine CBD-blend trial (Flint 2025). Milk allergen; bland, palatable. |
| Lemon balm extract | Chill | B | about 60 mg | Roy 2025 (20 beagles): calming effect under a mild stressor. Standardize to rosmarinic acid. |
| Vitamin E (d-alpha-tocopherol) | all | A | 8.1 IU/kg = about 150 IU | Placebo-controlled canine trial in atopic dogs (lower skin scores, higher antioxidant capacity). Doubles as the stabilizer for CBD, CBDA and the oil phase. |
| Astaxanthin (natural) | Mobility, Daily | B | about 5 mg | Beagle study: immune markers up, CRP down. Fat-soluble, goes in the oil phase. |
| Hyaluronic acid, oral (Mobilee) | Mobility | B (biomarker) | 27 mg | 55-dog post-surgical trial: synovial markers improved; no lameness outcome. |
| Bacillus coagulans GBI-30 spores | Daily | B | 1 billion CFU | Several small canine trials on stool quality; the one probiotic form that survives a low-water chew. Lactobacillus does not. Purina's BL999 is not available to third parties. Require end-of-shelf-life CFU on the COA. |
| Bovine colostrum | Daily | A/B | 0.1% of diet, about 300 mg | Purina trial: stronger vaccine response, higher gut IgA, more stable microbiome. Milk allergen. |
| Silybin phytosome (Siliphos) | Daily | B (PK) | 24 mg | Liver-support rationale: chronic CBD raises ALP in about half of dogs. Canine bioavailability shown; outcome data only inside Denamarin. Keep SAMe out of a moist chew (unstable). |
| PEA, ultramicronized | Mobility (itch) | C | 10โ15 mg/kg = 180โ270 mg | Open-label 160-dog itch study, no placebo; no canine arthritis trial. Shares the endocannabinoid pathway with CBD, so additive in theory, unproven in dogs. Self-affirmed GRAS in humans; same gray zone as every nutraceutical. Optional. |
| Boswellia, AKBA-standardized | Mobility | C | whole extract 400 mg per 10 kg (720 mg) or 50โ100 mg of a 20โ30% AKBA extract (extrapolated) | Open-label canine study (71% responders); label AKBA content is unreliable, so specify it on the COA. |
| Bioavailable curcumin (Meriva-type phospholipid complex) | Mobility | B (PK only) | 100โ200 mg curcuminoid-equivalent (extrapolated) | Beagle PK shows phospholipid and cyclodextrin forms absorb 2โ3 times plain curcumin; no canine outcome trial. Stains chews yellow. |
| Hydrolyzed collagen peptides (PETAGILE) | Mobility | B | per study weight band | 2024 placebo trial (n=31): force-plate gait improved. Complementary to UC-II. |
What the research rules out or caps
- Omega-3 at an arthritis dose does not fit in a chew. The effective canine dose is about 1,250 mg EPA+DHA a day; a 3.5 g chew's entire oil phase carries about 100 mg. Salmon or krill oil in the chew is a coat-and-palatability ingredient. Sell a separate fish oil if Mobility customers want the full dose. Specify GOED oxidation limits on any marine oil (peroxide value 5 or under, TOTOX 26 or under).
- MCT for cognition needs about 16 g a day; not chew-deliverable. MCT is a carrier choice only.
- Turkey tail has cancer-survival data at 100 mg/kg and a 2022 trial where adding it did not help. Not a wellness ingredient.
- Beta-glucan worked at 0.14% of diet (about 400 mg); a 100 mg chew dose is below the tested level.
- Melatonin: case series only; keep at or below 0.1 mg/kg (about 1.8 mg) or omit.
- Never: 5-HTP (serotonin syndrome, deaths), St John's wort (CYP inducer, interacts with trazodone and SSRIs), lavender or linalool (ASPCA-listed toxin), piperine (CYP and P-gp inhibitor, no canine data, would alter CBD clearance), xylitol, garlic, grapes. Cetyl myristoleate has no canine evidence.
Delivery and absorption (canine data)
- Lecithin plus sesame or MCT oil is the best-supported chew design: 25% sunflower lecithin in sesame oil held CBDA blood levels highest over two weeks, and a soft chew of the same extract matched the oils (Wakshlag 2020). Sunflower lecithin avoids the soy allergen.
- Liposomal CBD: 20 mg liposomal matched 50 mg plain in an arthritis trial (Verrico 2020), but liposomes are water dispersions and collapse in a dry chew. The practical equivalent is the lecithin-rich oil premix.
- Nano and water-soluble CBD: in a 32-dog study they absorbed no better than oil overall, only faster; the baked treat absorbed worst (35% of oil). Another reason for cold forming.
- Giving with food: canine data are sparse and conflicting; don't claim it.
- Cold-formed chew: no heat, no added water, water activity 0.6 or below. Protects CBDA, lecithin, astaxanthin, vitamin E and probiotic spores.
Quality standards a premium brand can truthfully claim
| Claim | What it takes |
|---|---|
| NASC Quality Seal | Membership, written QC manual to NASC cGMP, audit every 2 years, adverse-event reporting, random product testing, a substantiation file for every claim. Website and social count as labeling. |
| Made in an FDA-registered facility under 21 CFR 507 cGMP | Facility registration plus a written food-safety plan with hazard analysis. Never "FDA-approved." |
| Human-grade | Every ingredient and the finished product handled under human-food cGMP, in a facility registered as both human and animal food. Most chew plants fail this on facility status. Do not claim unless the manufacturer qualifies. |
| 3rd-party tested, ISO 17025 | Per-batch potency (CBD, CBDA, THC, THCA, CBG, CBGA), pesticides, heavy metals, residual solvents, microbials, mycotoxins, water activity, from an accredited lab with a proficiency-test record. |
| Label accuracy | Content uniformity on 10 chews per batch, within 10% of label. The Cornell survey found only 10 of 29 pet hemp products within 10%, and lead above oral limits in one. |
| Expiration date | Real-time and accelerated stability in the market pouch tracking CBD and CBDA separately, oxidation, microbes and CFU. |
| Allergens | Declare milk (casozepine, colostrum), egg (NEM), fish (oils), soy (if soy lecithin). |
| Prop 65 | Delta-9 THC is listed with no safe-harbor level, so any detectable THC sold into California needs the warning. Non-detect inputs sidestep it; lead in botanicals is the other trigger. |
| Vet formulated | No legal definition; an FTC express claim. A licensed vet must materially design the formula and the engagement record must exist. |
| U.S. Hemp Authority | FoodChain ID audit; Charlotte's Web and Pet Releaf hold it for pet lines. |
Drug interactions for the label and the vet conversation
- Chronic CBD raises ALP in about half of dogs at 4โ10 mg/kg/day (bone isoform); recommend liver values be checked on long-term use.
- Phenobarbital: a 9-beagle study found no change in phenobarbital kinetics with CBD.
- Trazodone: CBD did not add to trazodone's effect in the fireworks model; possible interaction flagged.
- Tramadol: CBD and CBDA inhibit its metabolism in canine liver microsomes (in vitro).
- NSAIDs: no in-vivo canine interaction study; Gamble 2018 allowed NSAIDs alongside CBD; ALP rose more with NSAID in Talsma 2024.
- Omega-3 plus NSAIDs or clopidogrel: additive platelet caution.
